What Are the Limitations of Single Plex Immunohistochemistry?
Despite its advantages, single plex IHC has some limitations:
- Antibody Specificity: Non-specific binding or cross-reactivity can lead to misleading results. - Sensitivity: The detection sensitivity may not be sufficient for low-abundance antigens. - Subjectivity: Interpretation of staining patterns can be subjective and requires expertise. - Limited Multiplexing: Only one antigen can be detected per assay, which may not be sufficient for complex studies.
