While highly sensitive and specific, RT-PCR does have some limitations:
- RNA Quality: The quality of RNA extracted from tissue samples must be high. Degraded RNA can lead to inaccurate results. - Quantification Challenges: Accurate quantification can be difficult due to variations in amplification efficiency. - Complexity of Tissue Samples: Heterogeneous tissues can complicate the analysis, as different cell types may have varying gene expression profiles.
