Despite its advantages, myelin staining has some limitations:
Artifacts: Improper fixation or staining can introduce artifacts that may complicate interpretation. Non-quantitative: Most myelin staining techniques are qualitative and may not provide precise quantitative data on myelin content. Resolution: Light microscopy techniques may not offer the resolution needed to study fine details of myelin structure.
