Despite its advantages, fluorometry has some limitations:
Photobleaching: Prolonged exposure to light can lead to the degradation of fluorophores, reducing fluorescence intensity. Autofluorescence: Some biological tissues exhibit natural fluorescence, which can interfere with the detection of specific fluorophores. Background Noise: Non-specific binding of fluorescent probes can generate background noise, affecting the accuracy of measurements.
