Despite its advantages, the Feulgen reaction has some limitations:
- Technical Sensitivity: The hydrolysis step must be carefully controlled. Over- or under-hydrolysis can lead to inaccurate results. - Not Suitable for RNA: Since the Feulgen reaction is specific to DNA, it cannot be used to study RNA content. - Time-Consuming: The multiple steps involved in the process make it relatively time-consuming compared to other staining techniques.
