One of the primary challenges in using fresh tissues is their rapid degradation. Enzymatic and microbial activities can quickly alter the tissue's composition. Therefore, timing and environmental control (temperature and humidity) are critical. Additionally, fresh tissues are more challenging to section and stain compared to fixed tissues, as they lack the rigidity provided by chemical fixatives.
