How does scATAC-seq work?

The scATAC-seq technique involves the use of a hyperactive Tn5 transposase that simultaneously cuts and tags accessible regions of chromatin with sequencing adapters. These tagged DNA fragments are then amplified and sequenced to identify regions of open chromatin, indicative of active regulatory elements such as enhancers and promoters. The single-cell resolution is achieved by isolating individual cells and barcoding their DNA fragments before sequencing.
