LHON is caused by mutations in the mitochondrial DNA, specifically in genes encoding components of the electron transport chain. The most common mutations are found in the ND1, ND4, and ND6 genes. These mutations impair the function of complex I of the respiratory chain, leading to decreased ATP production and increased oxidative stress in the retinal ganglion cells.
