The activation of NICD is initiated when a Notch receptor on the cell membrane binds to a ligand, such as Delta or Jagged, from an adjacent cell. This binding triggers a sequence of proteolytic cleavages. The final cleavage, mediated by the γ-secretase complex, releases NICD from the membrane, allowing it to translocate to the nucleus.
