Involucrin is synthesized in the cytoplasm of keratinocytes as these cells begin to differentiate. The gene encoding involucrin is regulated by various transcription factors, including AP-1 and Sp1. Upon synthesis, involucrin remains in the cytoplasm until the keratinocytes reach the upper layers of the epidermis, where it gets cross-linked by transglutaminases to form part of the cornified envelope.
