Pemetrexed inhibits several key enzymes involved in the folate pathway, including thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyltransferase. By blocking these enzymes, pemetrexed disrupts the production of nucleotides necessary for DNA and RNA synthesis. This results in the inhibition of cell division and ultimately leads to cell death.
