Glycogen synthase is primarily located in the cytoplasm of cells, particularly in the liver and skeletal muscle tissues. In histological sections, glycogen can be visualized using special staining techniques such as the Periodic Acid-Schiff (PAS) stain, which stains glycogen and other polysaccharides in a magenta color. The presence of glycogen synthase can be inferred by the abundance of glycogen granules within the cytoplasm of these cells.
