What are the Limitations of X-ray Crystallography?
Despite its advantages, X-ray crystallography has some limitations:
Crystallization Difficulty: Some molecules are difficult or impossible to crystallize, which limits the applicability of the technique. Static Snapshot: The technique provides a static picture of the molecule, which may not capture its dynamic nature in a biological context. Phase Problem: Determining the phases of the diffracted waves is challenging and requires additional techniques or computational methods.
