To prepare RIPA buffer, combine the following reagents and adjust the pH to 7.4-8.0:
50 mM Tris-HCl 150 mM NaCl 1% NP-40 or Triton X-100 0.5% Sodium deoxycholate 0.1% SDS Add protease and phosphatase inhibitors just before use to prevent protein degradation.
Store the buffer at 4°C for short-term use or aliquot and freeze for long-term storage.
