A cryostat microtome operates by maintaining a very low temperature, typically around -20 to -30 degrees Celsius, which freezes the tissue sample. The frozen tissue is then cut into thin sections using a steel knife. These thin sections, often just a few micrometers thick, are then placed onto glass slides for further microscopic examination.
